I have two machines, Double Diamond Run and a Double 3x 4x 5x. Is there anyway to get enhanced reel spin, rollup, etc. sounds from either one of these?

DD Run Vision will give you enhanced sounds since it draws from the sound from the top box flash card.  Guessing you have the standard, non-Vision, version?

DBL 3x4x5x Pay = nope...your out of luck.

My DDR is not a Vision but a standard S2000, it does have the enhanced cpu if that matters.

The "Plain Jane" S2000 DDR did not have a DSV assigned or designed for it.
I think the S2000 version was the first time that game came out.
The S2000 Vision DDR platform has sounds up in the
top box computer (PCMCIA card) as BettorSlots pointed out.
Yoeddy posted a video of the Reel Touch DDR platform which of course has extra sounds.

I'm sure the later platform versions of that game in I-Game and AVP have extra sounds as well -
if IGT continued the model line...
I've just haven't thought to look for them yet.
